  • Director of In Vivo Pharmacology

    Description

    Fulcrum Therapeutics is pioneering small molecule therapeutic approaches to gene regulation in genetically defined diseases. The current opening offers the opportunity to lead in vivo pharmacology efforts and research programs in a fast-paced, innovative environment as part of the Biology team.

    This role entails leadership of the In Vivo Pharmacology team with a primary focus on advancing our monogenic neuromuscular disorders portfolio. The position will focus on design and execution of studies aimed at building pharmacologic relationships between drug exposure, target engagement, pharmacodynamic markers and disease gene expression in rodents and large animals. The ideal candidate will lead both an internal team of scientists and partner effectively with external CROs to execute in vivo studies. The candidate will also lead collaborations with our Scientific Founders, Advisory Board members, and key scientific leaders. They will lead projects and be responsible for progressing therapeutic candidates from early discovery to IND-enabling studies. We are looking for excellent candidates to join our multidisciplinary team in an exciting and dynamic R&D environment.

    Responsibilities

    • Work with the team to prioritize therapeutic target and disease selection.
    • Lead therapeutic program efforts from target validation through candidate selection, and collaborate with translational medicine colleagues to transition into Phase 1 clinical trials.
    • Utilize pharmacologic in vitro characterization data to progress therapeutic candidates into in vivo studies.
    • Design and manage preclinical in vivo pharmacology studies including efficacy, pharmacokinetic and pharmacodynamic endpoint and biomarker studies, as well as provide input into toxicology studies.
    • Develop robust methods to quantitate modulation of target engagement and gene expression. Experience with chromatin or histone modification preferred.
    • In partnership with the Translational Medicine team, identify relevant biomarkers for transition from animal studies to humans.
    • Provide input into the design of translational research studies focusing on patient stratification and biomarker identification.
    • Contribute to the writing of relevant research reports to be included in regulatory filings.
    • Ensure CROs are maintaining laboratory notebooks in compliance with Fulcrum Therapeutics policies.
    • Communicate scientific progress to management, advisors, and scientific founders.
    • Recruit, train, manage and retain high-value employees.

    Requirements

    • Ph.D. in pharmacology, neuroscience, genetics or related field with 8+ years of relevant working experience in drug discovery or translational medicine.
    • The ideal candidate will have at least 6+ years of biopharmaceutical industry experience with a focus in neurologic or neuromuscular diseases. Experience in genetically defined biomarker strategies preferred.
    • Up-to-date knowledge of gene regulation mechanisms and methods preferred.
    • Experience designing and managing preclinical in vivo pharmacology studies, both internally and via outsourcing with CROs.
    • A demonstrated track record of independent critical thinking and scientific achievement.
    • Experience leading projects and working with cross-functional teams.

  • Computational Biologist - Molecular Profiling

    Description

    We are seeking a highly motivated computational biologist (open to all experience levels) to join our drug discovery efforts at Fulcrum specifically around target and biomarker identification in rare genetic diseases. This individual will focus on understanding molecular profiling data and their relationships to human phenotypic information using bioinformatic and machine learning approaches. The individual will work as part of a tight-knit computational biology group that is matrixed across several different experimental and translational teams.

    Responsibilities

    • Analyze genome, epigenome, and transcriptome datasets
    • Contribute to experimental study design and data interpretation, visualization
    • Expand efforts in single cell technologies
    • Identify, generate, and curate new sources of data

    Qualifications

    • An advanced degree or significant experience in computational biology, bioinformatics, molecular biology, computer science/engineering or related areas
    • Significant experience in regulatory genomics and transcriptomics
    • Familiarity with systems biology, single cell genomics

  • Computational Biologist - Data Science

    Description

    We are seeking a highly motivated Computational Biologist – Data Science at the Scientist or Sr. Scientist level to join our drug discovery efforts at Fulcrum. This position will focus primarily on the analysis of transcriptomic and epigenomic NGS data generated from a panel of compound perturbations in multiple cellular contexts across multiple cell-types in conjunction with human or model organism phenotypic data as it relates to our therapeutic programs.

    Responsibilties

    • This position will focus on the development and application of statistical methods for the integrated analysis of large­scale molecular datasets.
    • Perform exploratory data analysis of very large datasets from varying sources, drawing inferences and insights to design or tune statistical models/learning algorithms for identifying genes predictive of phenotypes or clinical outcomes.
    • Evaluate fits, error metrics and model diagnostics to assess and improve model performance.
    • Perform data collection and curation.
    • Interpret analysis to a broad array of wet-lab scientists.

    Qualifications

    • A PhD degree in a relevant technical field (i.e. Bioinformatics, Computational Biology, Statistics, Computer Science, Engineering, Physics, Math)
    • Experience with a wide array of machine learning algorithms and data mining methods (i.e. time series analysis, state space models, adaptive filtering, mixed effect models, hierarchical Bayes, Markov models, decision trees, boosting, random forests, support vector machines).
    • Experience analyzing genomics, epigenetics and transcriptomic datasets.
    • Proficient programming skills (e.g. Python, C++, Scala).
    • Expert knowledge of a statistical analysis package (e.g. R, Matlab, SAS).

  • Scientist, Cell-based Screening and Target ID

    Description

    As a Scientist, you will be expected to work equally well in self-starting mode and also as a member of a matrixed team within Fulcrum. Your focus will be to develop, implement and execute cell-based assays using patient-derived cellular models, including neurons and myotubes, for high-throughput chemical and CRISPR screens. As one of the key hires for the company, you will work in close collaboration with the biology team and scientists to support our early-stage drug discovery pipeline.

    Responsibilties

    • Develop, optimize and troubleshoot cell-based assays in 384 well plate format.
    • Collaborate with developmental cell biologists to maintain high quality myotube and neuronal cell cultures.
    • Collaborate with information technologists to implement a screening pipeline to track samples from entry, processing to analysis.
    • Program, integrate assays and execute screens using state-of-the-art automation systems.
    • Prepare, order, and maintain stocks of necessary reagents and supplies.
    • Document, compile, analyze and present experimental data.
    • Generate and maintain clear experimental records.
    • Organize and clearly present experimental results to the project team.

    Qualifications

    • Ph.D. with at least 2+ years of experience or M.Sc. with 5+ years of relevant post-graduate and / or industry experience.
    • At least 3-5 years of hands-on experience in assay development and implementation, plate-based pharmacology and/or high throughput screening (small molecule and CRISPR).
    • Experience in culturing primary, neuronal or other differentiated cell types is desirable.
    • Strong background using common cell and molecular biology tools, such as RT-PCR, ELISA, western blotting, DNA/RNA prep and IHC.
    • Experience with gene editing (CRISPR Cas9) is a plus.
    • Experience with data analysis software and tools such as Genedata Screener, Spotfire or experience in NGS library prep and/or analysis is desirable.
  • Senior IT Infrastructure Engineer

    Description

    We are looking for a hands-on Senior IT Infrastructure Engineer to join our team that builds, maintains, and supports Fulcrum’s corporate IT infrastructure and internal staff. This role is an exciting opportunity to be a critical part of a team that is fueling a growing biotech company. In addition to having a solid technical background, this candidate will have excellent communications skills, be able to work with a managed services provider, and have a passion for service and all things IT. The ideal candidate will have previous experience with or have directly worked in the Life Sciences industry.

    Responsibilties

    The IT team is responsible for help desk support, server administration, IT security, workstations, laptops, network configuration, and Lab IT/Informatics support. It also is responsible for audio/video conference systems, VoIP, file storage systems, and backups. Additionally, the team supports and maintains Fulcrum’s cloud operations in AWS.

    Qualifications

  • Senior/Principal Biostatistician

    Description

    We are seeking an experienced Biostatistician to join the team and lead the statistical strategy within the Clinical Development function. This individual would report to the Head of Clinical Development and would be responsible for the biostatistical data management function. Additionally, they will provide statistical input and support to ensure plans and analyses are accurately designed, specified and conducted; and will ensure achievement of statistical deliverables with efficiency and quality.

    Responsibilities

    • Serve as biostatistics leader at Fulcrum and provide internal training to the team as needed.
    • Provide statistical oversight of all research study protocols in the preclinical and clinical space and participate in reviews as needed.
    • Ensure consistency in statistical methods applied to all study designs.
    • Provide detailed review of existing data to inform assumptions for design of clinical trials and to set appropriate assumptions for power calculations.
    • Provide sample size and design proposals for Clinical Trials.
    • Biostatistics project management including but not limited to vendor hiring, vendor management, directing statistical programming, and escalating issues in a timely manner.
    • Be responsible for oversight of data management in outsourced clinical trials.
    • Function as study level statistician for internal studies.
    • Ensure high quality, timely delivery of statistical analysis plans and study tables/listings and statistical reports.

  • Research Associate/Sr. RA

    Description

    We are seeking a highly motivated research associate to join our efforts in the Biology Department at Fulcrum to participate in building our discovery pipeline for CNS and neuro-muscular diseases. This individual will contribute to scientific efforts focused on building a relationship between the correction of gene expression profiles and functional improvement of disease phenotypes in complex human cellular models.

    Responsibilities

    • Design, develop and execute human iPSC-derived cell culture models of CNS and neuro-muscular diseases to identify functional phenotypes through a variety of physiologically relevant assay techniques. Cell culture models will be assessed utilizing the assay technologies of MEA electrophysiology, seahorse metabolic pathway assays, high-content imaging, fluorescent plate readers, and protein quantification.
    • Analyze complex data sets and clearly communicate results to the larger group.
    • Actively participate across functional groups and programs to provide knowledge of physiological, biochemical and pharmacologic techniques required to examine target expression, modulation and function.
    • Demonstrates understanding of theory and application of cell biology and physiology assays to examine gene expression, subcellular localization, electrophysiological function, fluorescent microscopy techniques and quantitative image analysis.

    Qualifications

    • BS/MS in biology, molecular and/or cellular biology
    • 2-5 years of hands-on experience in neurologic, muscle or cardiovascular diseases
    • Knowledge, application and analysis of compound pharmacology testing using in vitro disease models.
    • Experience performing and analyzing MEA and/or high definition MEA assays is desired, especially experience with specialized analysis software (i.e. MATLAB, python, or R).
    • Skilled in pharmacologic assessment of both small molecules and other therapeutic modalities in primary or stem cell derived cellular systems.
    • Proficiency in development of optical and image-based assay approaches and analysis.
    • Ability to identify and solve technical issues that arise with equipment, especially electrophysiology equipment.
    • Excellent written and oral communication as demonstrated in the quality of the employee’s verbal interactions on projects in a team environment.
    • Experience and effectiveness working in a multidisciplinary team environment with common goals and operating in a matrixed environment is essential.
    • Rigor, attention to detail and problem-solving skills are required.
    • Experience preferred in whole cell patch clamp techniques in either clonal or primary cultured cells, including neurons and myocytes.
